Whether the drug improves hard clinical outcomes is not settled. No completed trial has shown a reduction in heart attacks or strokes among treated patients, although a dedicated cardiovascular outcomes study has been discussed in the literature. Investigators have also examined hepatic fat in people with HIV and fatty liver disease, cognitive measures in small cohorts, and changes in bone density. Regulatory labeling emphasizes monitoring of insulin-like growth factor 1 because supraphysiologic levels raise questions about tissue growth, and the clinical significance of that signal remains an open question rather than a demonstrated harm.
Binding of tesamorelin to the growth hormone-releasing hormone receptor on anterior pituitary somatotrophs activates a Gs protein pathway, raises cyclic AMP, and triggers release of stored growth hormone into the bloodstream. Because the analogue resists dipeptidyl peptidase-4, its plasma residence time exceeds that of native GHRH, producing a larger and more sustained secretory signal. The released growth hormone then acts on the liver and peripheral tissues to raise insulin-like growth factor 1, which feeds back on the hypothalamus and pituitary. This axis explains both the intended effects on fat distribution and the biological markers used to track them.
Regulatory position depends on jurisdiction and on the form in which the material is sold. A branded product holds approval in the United States for a defined indication, and prescribing is confined to that label. Material marketed for laboratory research is not evaluated for human use and carries no such clearance. Independent verification therefore rests on certificates of analysis, third-party testing, and documented chain of custody. The substance also appears on the World Anti-Doping Agency prohibited list within the category covering growth hormone-releasing factors.
Lyophilized material is typically held under refrigeration between two and eight degrees Celsius, shielded from light and ambient moisture. Peptides of this size adsorb to glass and plastic, so working procedures often call for low-binding containers and as few transfers as possible. Absorbed water during weighing shifts the apparent mass of a sample, and controlling room humidity reduces that source of error. Once dissolved, solutions are kept cold and used within the interval printed on the accompanying label or certificate. Degradation accelerates markedly in dilute aqueous form.
Identity and purity are judged through a combination of chromatographic and mass spectrometric techniques. Reversed-phase high-performance liquid chromatography separates the intact peptide from truncated, oxidized, and deamidated variants, and the resulting peak-area percentages yield a purity figure. Electrospray ionization mass spectrometry confirms the expected molecular mass and can expose unanticipated modifications. Amino acid analysis and peptide mapping support sequence fidelity, while water content, pH, sterility, and bacterial endotoxin testing describe the physical and microbiological attributes of a finished lot.

Measuring the effect of a growth hormone-releasing hormone analogue requires markers that reflect pituitary output rather than the peptide itself. The two most frequently used are growth hormone and insulin-like growth factor 1. Growth hormone fluctuates sharply across the day and responds to sleep, stress, and meals, so isolated readings can be difficult to interpret. Insulin-like growth factor 1 changes more slowly and is often treated as the more stable integrated marker of axis activity.
Because growth hormone is released in pulses, single measurements can misrepresent overall secretion. Investigators sometimes use repeated sampling or overnight profiles to capture the pattern rather than a single value. Provocative testing, in which a stimulus is given and the response is tracked over time, offers another way to characterize the axis. Each approach carries trade-offs between sensitivity, burden on the participant, and the influence of non-target variables.
Insulin-like growth factor 1 is produced largely in the liver in response to growth hormone signaling. Its concentration shifts over days rather than minutes, which makes it practical for tracking changes across a study period. Interpretation still depends on age, nutritional status, and concurrent illness, all of which independently affect the marker. Reference ranges are therefore stratified, and comparisons are usually made within an individual over time rather than against a single population threshold.
Lotte Bjerre Knudsen (born 10 March 1964) is a Danish scientist and university professor. She led the development of liraglutide and oversaw the development of semaglutide, two notable drugs approved for indications in the treatment of diabetes and obesity.
== Use as an educational tool == The AIDA software comes with forty educational case scenarios as standard, each of which represents a 'snapshot' of the metabolic status of a typical person with respect to insulin-dependent type 1 diabetes mellitus. It is easy for users to add or create further case scenarios, as required. Examples of the application of AIDA as an educational tool can be found in various published journal articles.
In Konsumentombudsmannen v Gourmet AB the Court suggested that a total ban for advertising alcohol on the radio, TV and in magazines could fall within article 34 where advertising was the only way for sellers to overcome consumers' "traditional social practices and to local habits and customs" to buy their products, but again the national courts would decide whether it was justified under article 36 to protect public health. Under the Unfair Commercial Practices Directive, the EU harmonised restrictions on restrictions on marketing and advertising, to forbid conduct that distorts average consumer behaviour, is misleading or aggressive, and sets out a list of examples that count as unfair. Increasingly, states have to give mutual recognition to each other's standards of regulation, while the EU has attempted to harmonise minimum ideals of best practice. The attempt to raise standards is hoped to avoid a regulatory "race to the bottom", while allowing consumers access to goods from around the continent.
In June 2024, a third Arizona location opened in Goodyear. White Castle announced on November 25, 2019, that the chain would return to Florida after previously leaving the state in 1968, with plans to open the first restaurant in Orlando. A ghost kitchen, operated out of the restaurant while it was under construction, overloaded Uber Eats when it opened for one day on February 24, 2021. The Orlando location opened on May 3, 2021. It is the world's largest White Castle, located on Daryl Carter Parkway off Interstate 4. The opening coincided with White Castle's 100th anniversary. In 2020, White Castle began testing an automated cooking robot called Flippy in a number of its Chicago-area stores, and then equipped a larger number of locations with the updated Flippy 2 model in November 2021. The system is able to discriminate amongst burgers, chicken fingers, and french fries, pick them up, cook them through automated temperature detection and flipping action, place the cooked item in a fry basket, and in turn place the basketed food in an area for holding hot items. The Flippy 2 model can operate without human intervention and produce 60 baskets of food per hour. By the end of 2022, a third of White Castle locations were expected to be equipped with the device. In August 2025, White Castle announced it would open its first location in Texas in the northern Dallas suburb of The Colony. The restaurant is scheduled to open in 2026 in a mega retail and restaurant development known as Grandscape.
== Early life and education == Susan Epstein (later changed to Leeman by marriage) was born on May 9, 1930, in Chicago, Illinois. Her mother was born in the United States and her father had emigrated from Russia to New York City. Her father was an academic metallurgist and her mother attended college at George Washington University at a time when few other women did. Susan also had one older brother named Henry. When Susan was six weeks old she and her family moved to Columbus, Ohio, and then to Bethlehem, Pennsylvania when she was six years old. There she grew up a part of a middle class Jewish family. She often faced discrimination in the form of antisemitism and sexism as she pursued a career in science. During her childhood Leeman attended Hebrew School and was a Girl Scout. She decided to attend Goucher College, which was an all-girls’ school at the time, from which she received a bachelor's degree in physiology in 1951. She then applied to and was accepted by Harvard Medical School, but her academic program was administered through Radcliffe College. Thus, Radcliffe College was where she received her master's degree and PhD from in 1954 and 1958 respectively. Leeman was the only woman in her class to make it through the graduate program and continue a career in science. During her time in graduate school she was introduced to the field of neuroendocrinology, within which she was able to explore her passion for how the mind connects to the body.

Others have found that no bias exists on average in an officer's decision to stop a citizen, but bias may exist in an officer's decision to frisk a citizen. Carter oversaw the largest scale back of incarceration in US history. He presided over a 34% decline in sentenced prisoners between 1977 and 1980, a reduction of 9,625 inmates. That was the largest of any president on record, both as a percentage and in absolute numbers.
Drew won an athletics scholarship to Amherst College in Massachusetts, where he played on the football as well as the track and field teams, and graduated in 1926. After college, Drew spent two years (1926–1928) as a professor of chemistry and biology, the first athletic director, and a football coach at the historically black private Morgan College in Baltimore, Maryland, to earn the money to pay for medical school. For his medical career Drew applied to Howard University, Harvard Medical School, and later McGill University. Drew lacked some prerequisites for Howard University, and Harvard wanted to defer him a year, so to begin medical school promptly, Drew decided to attend McGill's medical school in Montreal, Canada. It was during this stage in his medical journey that Drew worked with John Beattie, who was conducting research regarding the potential correlations between blood transfusions and shock therapy. Shock occurs as the amount of blood in the body rapidly declines which can be due to a variety of factors such as a wound or dehydration. As the body goes into shock, both blood pressure and body temperature decrease which then causes a lack of blood flow and a loss of oxygen in the body's tissues and cells. Eventually, it became clear that transfusions were the solution to treating victims of shock, but at the time there was no successful method of transportation or mass storage of blood, leaving transfusions to be extremely limited to location.
== Tetramers in immunology == In immunology, MHC tetramers can be used in tetramer assays, to quantify numbers of antigen-specific T cells (especially CD8+ T cells). MHC tetramers are based on recombinant class I molecules that, through the action of bacterial BirA, have been biotinylated. These molecules are folded with the peptide of interest and β2M and tetramerized by a fluorescently labeled streptavidin. (Streptavidin binds to four biotins per molecule.) This tetramer reagent will specifically label T cells that express T cell receptors that are specific for a given peptide-MHC complex. For example, a Kb/FAPGNYPAL tetramer will specifically bind to Sendai virus specific cytotoxic T cell in a C57BL/6 mouse. Antigen specific responses can be measured as CD8+, tetramer+ T cells as a fraction of all CD8+ lymphocytes. The reason for using a tetramer, as opposed to a single labeled MHC class I molecule is that the tetrahedral tetramers can bind to three TCRs at once, allowing specific binding in spite of the low (1 micromolar) affinity of the typical class I-peptide-TCR interaction. MHC class II tetramers can also be made, although these are more difficult to work with practically.
Maúrtua further added that Castillo met Maduro during the VI Summit of CELAC, where many Latin American leaders were present. The Foreign Minister stated that in such multilateral events, spontaneous meetings are natural. Subsequently, the Free Peru caucus accused Maúrtua of trying to impose his political agenda on the President, and called for Maúrtua's resignation: "The foreign minister remained silent... in the face of the audacity of his Vice Foreign Minister Luis Enrique Chávez to contradict the president in his decision to maintain friendly relations and cooperation with Venezuela and the Maduro government. Therefore, having transgressed the provisions of Article 118, paragraph 11 of the Political Constitution, our caucus demands the resignation of both the minister and the vice minister." On 15 October 2021, following the appointment of the Vásquez cabinet, Richard Rojas Garcia was appointed as Peru's ambassador to Venezuela. This in effect established official diplomatic relations with Venezuela. Some media outlets have claimed that Rojas is the right-hand man of Vladimir Cerrón. However, soon afterward a judge ruled that Rojas could not leave the country for 6 months, due to his alleged link with members of "Los Dinámicos del Centro." Afterward, Foreign Minister Óscar Maúrtua claimed that Rojas' appointment as ambassador to Venezuela never took place, "since the corresponding process was not concluded. In this sense, Peru is proceeding to inform the Government of Venezuela that the proposed appointment of Mr.
== Pharmacology == In tests with rabbits, a significant improvement in protection against arrhythmic effects and infarct size reduction was observed after administrating exogenously SRTX-c (in dosage of 0.24 nmol/kg, i.v.) prior the coronary occlusion accident. That was achieved thanks to the ability of SRTX-c to activate selected ETB receptors. In rat thoracic aorta, the contractile activity is grouped as follows: ET-1 > SRTX-b > SRTX-a > SRTX-c at lower concentrations, but SRTX-b > ET-1 > SRTX-a > SRTX-c at higher concentrations. Intra-arterial injections of SRTX-b cause a dose-dependent increase in perfusion pressure at doses ranging from 30 to 300 pmol. The vasoconstrictor activity of SRTX-b is less remarkable than that of ET-1 at doses lower than 100 pmol, while at a dose of 300 pmol the activity of SRTX-b is greater than that of ET-1. The time required for the recovery of perfusion pressure to baselines after a bolus injection of 300 pmol SRTX-b is shorter than that of ET-1. The threshold vasoconstrictor dose of SRTX-a is 3 times larger than that of SRTX-b. At a dose of 300 pmol, the rise in perfusion pressure due to SRTX-a is about 8 times smaller than that of SRTX-b. SRTX-c exhibits a feeble vasoconstriction producing a very small increase in perfusion pressure.

The existence of deuterium at a low but constant primordial fraction in all hydrogen is another one of the arguments in favor of the Big Bang over the Steady State theory of the Universe. The observed ratios of hydrogen to helium to deuterium in the universe are difficult to explain except with a Big Bang model. It is estimated that the abundances of deuterium have not evolved significantly since their production about 13.8 billion years ago. Measurements of Milky Way galactic deuterium from ultraviolet spectral analysis show a ratio of as much as 23 atoms of deuterium per million hydrogen atoms in undisturbed gas clouds, which is only 15% below the WMAP estimated primordial ratio of about 27 atoms per million from the Big Bang. This has been interpreted to mean that less deuterium has been destroyed in star formation in the Milky Way galaxy than expected, or perhaps deuterium has been replenished by a large in-fall of primordial hydrogen from outside the galaxy. In space a few hundred light years from the Sun, deuterium abundance is only 15 atoms per million, but this value is presumably influenced by differential adsorption of deuterium onto carbon dust grains in interstellar space. The abundance of deuterium in Jupiter's atmosphere has been directly measured by the Galileo space probe as 26 atoms per million hydrogen atoms. ISO-SWS observations find 22 atoms per million hydrogen atoms in Jupiter, and this abundance is thought to represent close to the primordial Solar System ratio.
Californium-252 (Cf-252, 252Cf) undergoes spontaneous fission with a branching ratio of 3.09% and is used in small neutron sources. Fission neutrons have an energy range of 0 to 13 MeV with a mean value of 2.3 MeV and a most probable value of 1 MeV. This isotope produces high neutron emissions and has a number of uses in industries such as nuclear energy, medicine, and petrochemical exploration.

=== Insulin interactions === D-chiro-inositol is known to be an important secondary messenger in insulin signal transduction. It accelerates the dephosphorylation of glycogen synthase and pyruvate dehydrogenase, rate limiting enzymes of non-oxidative and oxidative glucose disposal. Exogenous DCI may act to bypass defective normal epimerization of myo-inositol to DCI associated with insulin resistance and at least partially restore insulin sensitivity and glucose disposal. In clinical practice, it improves insulin signaling, thus restoring physiological insulin levels in resistant subjects.

It mimics a natural hypothalamic signal that tells the pituitary to release growth hormone. The result is a rise in circulating growth hormone and, indirectly, in insulin-like growth factor 1. Over weeks of treatment this shift is associated with a selective decrease in fat stored inside the abdomen.
The primary measure is usually a cross-sectional abdominal scan that separates internal fat from fat just under the skin. Waist circumference and body weight are recorded as secondary measures because they are easy to obtain but do not distinguish the two fat compartments. Hormone and metabolic blood tests are collected alongside the imaging.
Available follow-up data indicate that visceral fat drifts back toward pretreatment levels once injections stop. The change is therefore best described as treatment-dependent rather than permanent. Investigators continue to debate whether intermittent or repeated courses would preserve any benefit.
Reconstituted solutions are kept cold and used within the period stated on the label or certificate. Repeated warming and cooling cycles should be avoided because they encourage aggregation and gradual loss of potency.
